Inheritance: Knowlton (1929) provided evidence of autosomal recessive inheritance in Rhode Island Red chickens. Shiina and Kobayashi (2024) reported a sex linked mode of inheritance in Silkie Fowl chickens.

Mapping: Okumura et al. (2013) conducted a genome scan with 122 informative miccrosatellites on 88 Rhode Island Red chickens from several generations of families segregation for this disorder. Linkage analysis highlighted just one region near microsatellite MCW0332 on chromosome GGA12. Subsequent homozygosity mapping narrowed the region "to approximately 1.1 Mb, between GGA12_28 and GGA12_30", which contains at least 10 genes. Shiina and Kobayashi (2024): "Normal and congenital loco-affected Silkie Fowl chicks exhibited no differences in the frequencies of markers on chromosome 12 adjacent to the congenital loco locus reported in a previous study in Rhode Island Red chickens. ... Bulked segregant analyses using next-generation sequencing narrowed the causative region of congenital loco to approximately 3.3 Mb between bases 9,569,012 and 12,863,792 on chromosome Z."
